プレーヤーは、映画「天使と悪魔」の主人公ロバート・ラングドン教授となって制限時間内にパズルを解き、誘拐された枢機卿を救い出しつつバチカン市国に隠された爆弾を見つけ出す必要がある。映画のストーリーを楽しめるストーリーモード、60種類以上のパズルなどが用意されている。
